Detailed explanation of the Bovine Corneal Opacity and Permeability Test (BCOP) required for Amazonian Eye Cosmetics

What is BCOP certification?

The bovine corneal opacity and permeability test (BCOP) is an in vitro eye irritation test developed by Gautheron et al. (1992). This test evaluates the eye irritation potential of the test article using fresh bovine cornea obtained from a slaughterhouse. The type of damage caused by the exposure of the test article was quantitatively detected by the changes in the turbidity of the cornea and the permeability to fluorescein. The BCOP test can be used to study the mechanisms that lead to injury. Protein degeneration or stromal swelling can lead to corneal opacity, while impaired corneal barrier function and cell membrane junctions between corneal epithelial cells can manifest as changes in corneal permeability. In addition, histological endpoints can be supplemented to assess corneal swelling, dehydration, or morphological changes. Histological evaluation is mainly to evaluate the type of damage observed and the depth of damage to the cornea. Methods The chicken embryo allantoic membrane vascular test (CAMVA), the bovine corneal opacity permeability test (BCOP) and the Draize rabbit eye test were used to test 60 kinds of cosmetics, and to predict the classification of eye irritation. Results 41 non-irritating samples and 18 irritating samples could be distinguished by CAMVA method. 35 non-irritant, 21 mild-moderate irritant and 4 severe irritant samples were predicted by BCOP method. The combined CAMVA-BCOP method can significantly improve the discrimination ability of stimulus degree, and the consistency with the in vivo experimental classification is 98.3%. Conclusion The integrated test strategy of combined CAMVA-BCOP can be used for the eye irritation evaluation of cosmetic products, and its prediction range can cover non-irritant to severe irritation.Mainly to test the irritation of the product to the eyes

Test model: bull's eye horn tissue (by-product from slaughterhouse)

End Points:

1 Turbidity value, determined by measuring the amount of light transmitted through the cornea

2 Permeability, determined by measuring the amount of fluorescein dye that penetrates the cornea

Positive and negative controls were set for each experiment.

Sensitivity

BCOP is primarily used to evaluate test articles for moderate to severe eye irritation/corrosion. However, the exposure protocol can be modified to increase the sensitivity of the test system for the evaluation of mild eye irritation/corrosive formulation components.

Test object compatibility

This test can be used for water-soluble preparations as well as insoluble preparations; it can test solid test substances and liquid test substances. Since the test article is administered topically, the test system is also suitable for emulsions, pastes, highly viscous substances and powders.

Alternative endpoints

BCOP has multiple endpoints for which mechanistic studies can be conducted. In addition, histological endpoints can be supplemented to assess the type and depth of damage observed. Based on the work of Maurer and Jester (2002), the depth and extent of injury can be used to predict the severity and outcome of corneal injury. When necessary, test article concentrations and exposure times can be adjusted to suit specific physical properties or customer needs.

validity

In the US and Europe, the BCOP has been approved for the identification of corrosive substances and for the identification of chemicals not required to be classified under UN GHS, the OECD guideline (TG437) was adopted in 2009 and was revised in 2013 and accepted by the US EPA The BCOP test was used as one of the grading strategies for labeling antibacterial claims on cleaning products.

Histology

Depending on the purpose of the study, the results of the histological evaluation may be used in a number of ways. Histological results can be used to confirm corneal irritation levels predicted by turbidity values and permeability. In addition, it can be used to illustrate the type and depth of corneal damage caused by the test article.

BCOP test process

limitation

The limitation of the BCOP method is that it does not consider the stimulating effect of the test substance on the iris and conjunctiva. Some substances may damage the conjunctiva, but the effect on the cornea is not obvious. If only the BCOP method is used, it may only reflect the cornea of the compound. damage to the conjunctiva while ignoring possible effects on the conjunctiva.

What products need this certification?

Such as eye shadow, liquid eyeliner, magnetic eyelashes, false eyelashes, etc. may cause certain irritation to the eyes, so Amazon is now strictly checking these relevant certifications
